Terrein Inhibits Aggressive Phenotype of A549 Human Lung Cancer Cell through Suppression of HIF-1α

چکیده

Terrein is a fungal metabolite that has already been reported with anticancer properties. However, the effect on aggressive phenotype of cancer cells not elucidated yet. In present study, cytotoxicity terrein was first determined against lung (A549) model and compared several normal cell lines (Vero, L6, H9C2 cells). The data demonstrated had specific A549 relative to high selectivity index values. Then, hypoxic recognized induce abilities established in by cobalt chloride (CoCl2) stimulation. With this model, could reduce HIF-1α, marker hypoxia, inhibit both migration invasion which more explicit. Our results potential new role as anti-aggressive inhibiting through HIF-1α reduction. HIGHLIGHTS Terrein, secondary bioactive extracted from Aspergillus terreus, demonstrates less cytotoxic CoCl2 treatment successfully used for creating resulting HIF-1a augmentation enhancement expression invasive ability anti-metastatic agent GRAPHICAL ABSTRACT

Digoxin Downregulates NDRG1 and VEGF through the Inhibition of HIF-1α under Hypoxic Conditions in Human Lung Adenocarcinoma A549 Cells

Digoxin, an inhibitor of Na+/K+ ATPase, has been used in the treatment of heart-related diseases (such as congestive heart failure and atrial arrhythmia) for decades. Recently, it was reported that digoxin is also an effective HIF-1α inhibitor. We investigated whether digoxin could suppress tumor cell growth through HIF-1α in non-small cell lung cancer cells (A549 cells) under hypoxic condition...

Daxx inhibits hypoxia-induced lung cancer cell metastasis by suppressing the HIF-1α/HDAC1/Slug axis

Hypoxia is a major driving force of cancer invasion and metastasis. Here we show that death domain-associated protein (Daxx) acts to negatively regulate hypoxia-induced cell dissemination and invasion by inhibiting the HIF-1α/HDAC1/Slug pathway. Daxx directly binds to the DNA-binding domain of Slug, impeding histone deacetylase 1 (HDAC1) recruitment and antagonizing Slug E-box binding. This, in...

Psoralidin induces autophagy through ROS generation which inhibits the proliferation of human lung cancer A549 cells

Psoralidin (PSO), a natural furanocoumarin, is isolated from Psoralea corylifolia L. possessing anti-cancer properties. However, the mechanisms of its effects remain unclear. Herein, we investigated its anti-proliferative effect and potential approaches of action on human lung cancer A549 cells. Cell proliferation and death were measured by MTT and LDH assay respectively. Apoptosis was detected...

15-LOX-1 suppression of hypoxia-induced metastatic phenotype and HIF-1α expression in human colon cancer cells

The expression of 15-lipoxygenase-1 (15-LOX-1) is downregulated in colon cancer and other major cancers, and 15-LOX-1 reexpression in cancer cells suppresses colonic tumorigenesis. Various lines of evidence indicate that 15-LOX-1 expression suppresses premetastatic stages of colonic tumorigenesis; nevertheless, the role of 15-LOX-1 loss of expression in cancer epithelial cells in metastases con...

ATM induces radioresistance of non-small cell lung cancer A549 cells by downregulation of MDMX

Background: Tumor radioresistance leads to a reduction in the efficiency of radiation therapy. It is very important to explore the cellular mechanisms leading to radioresistance and to find potential therapeutic targets, which might improve the efficacy of radiation therapy. This study was to investigate the role of ataxia-telangiectasia mutated (ATM) and murine double minute X (MDMX) in radior...
